Abstract: Cinnamyl Alcohol Dehydrogenase (CAD) is a candidate gene influencing lignin content and composition in vascular plants. Sequence variation in CAD may be responsible for changes in lignin formation which affect wood properties. In this study, two important species in Queensland Forestry plantations, Pinus elliottii var. elliottii and Pinus caribaea var. hondurensis were examined for sequence variation in CAD. The individuals tested were parents of interspecific hybrid breeding populations. Polymorphism in CAD was detected and mapped in one population and the potential to map CAD in another two populations was established. We used an efficient and cost effective method for SNP detection and population screening involving single strand conformation polymorphism (SSCP) analysis with 5'- tail labelling of PCR products
